Fluorescence microscopy movie shows seeds (labeled with Alexa 568), elongated MTs (labeled with Alexa 647) and Neutravidin beads (with DyLight 488). Each filament is attached to a single bead at the biotinylated seed region. Subsequently, the bottom MT is moved closer to the top one, and depletion-induced bundle spontaneously forms. Overlap region can be distinguished by increased brightness. Bottom MT is then moved up, thus changing the MT overlap.
